Uchiha Yuki knew that Senju Wakama was one of the sons of the Senju clan leader.
During his brief conversation with Uchiha Madara, Senju Wakama had mentioned "Tsukuyo," who was clearly his younger sister.
This meant that Tsukuyo was the Senju princess.
The teasing tone in Senju Wakama's voice hinted at some ambiguous relationship between Uchiha Madara and the Senju princess.

Senju Wakama wasn't as tsundere as his second brother, Tobirama. He admitted frankly, "I can't quite describe the feeling, but I'm definitely confused and a bit uncomfortable."
